Induction of neuronal dopaminergic differentiation by non-Wnt related small molecules

Stem cell based HTS analysis of newly synthesized substances - application of cell lines for the characterisation of small molecules inducing dopaminergic stem cell differentiation (link to SFB).
HTS of small molecule libraries is performed for modulation of transcriptional activity in ReNcells VM, NeuroProgen and SA002.5 cells using eGFP reporter gene constructs coupled to the following promoters: nuclear receptor Nurr1, fibroblast growth factor FGF8 and paired-like homeodomain transcription factor (pituitary homeobox) PiTX3.Subsequently, active compounds are tested in a high content setting for their influence on marker expression for NPCs, astroglial cells and neurons.
